Known natural hormones as well as relatively unknown environmental pollutants seem to have the ability to potentially disrupt the endocrine system of man and animal in such a way that harmful effects on their development and reproduction can occur. \r\nThe first indications of possible effects of endocrine disrupting substances and the presence of these substances in the Scheldt estuary have recently been published. The industrial areas of Ghent and Antwerp are to a large extent responsible for this pollution. Therefore, there is an obvious need to investigate the occurrence of endocrine disruption in the Scheldt estuary. In addition, a detailed knowledge of the distribution and long-term effects of these substances is needed in the framework of future-oriented policy measures at the national and European level.\r\n\r\n\r\n<li>Project description \r\n\r\n<ul><li>Objectives\r\n\r\nThe major objectives within the project are:\r\n\r\n- analyses of endocrine disrupters in water, suspended solids, sediment and biota (mysids, gobies);\r\n- in vitro evaluation of the estrogenic and androgenic potency of samples of water, suspended solids and sediment;\r\n- ecotoxicological and in situ evaluation of the effects on the resident mysid populations (short, intermediate and long-term);\r\n- risk assessment of endocrine disrupters in the Scheldt estuary. \r\n\r\n\r\n<li>Methodology\r\n\r\nThe proposed project can be divided into four different research phases:\r\n\r\nPhase I: Distribution of endocrine disrupting substances in the Scheldt estuary (chemical analyses, analyses on biota, in vitro analyses)\r\n\r\nThe Scheldt estuary will be sampled three times a year (spring, summer and winter) for the duration of the project. On each sampling date, sediment, water, suspended solid and biota (mysid shrimp and gobies) will be collected. An extended list of natural hormones and endocrine disrupting chemicals will be measured in these samples. \r\n\r\nThe different water, sediment and suspended solid extracts will be tested in vitro to assess their potential to bind with the (human) estrogen and androgen receptor. The results of these analyses will allow an identification of the estrogenic and androgenic potency in different environmental matrices of the Scheldt estuary.\r\n\r\nPhase II: Evaluation of the exposure of biota from the Scheldt estuary to endocrine disrupting substances\r\n\r\nBased on the results of the first sampling campaigns priority substances will be selected. These priority endocrine-disrupting chemicals will be used in laboratory experiments with mysid shrimp to assess acute and chronic effects (Phase III).\r\n\r\nPhase III: Ecotoxicological evaluation of the effects of endocrine disrupting substances occurring in the Scheldt estuary on resident mysid shrimp populations (laboratory and field studies)\r\n\r\nAcute and chronic laboratory tests with mysid shrimps\r\nTo evaluate the possible effects of the pollutants (retained in phase II) on mysid shrimp, acute and chronic test will be performed in the laboratory. Mysid shrimp will also be exposed in multi-generation experiments during two generations (until the first generation of juveniles produces juveniles again) to assess the effects of endocrine disrupters during prolonged exposure. \r\n\r\nPopulation study of the native mysid shrimp in the Scheldt estuary\r\nThe mysid shrimp population of the Scheldt estuary will be sampled and studied during one year. In this way, the present population dynamics can be compared with the available historical data and possible changes can be detected. In addition, a more in-depth field study on the possible endocrine disruption effects will be conducted based on the results of the laboratory toxicity experiments. Sensitive endpoints identified in the laboratory experiments will be validated in situ on the resident mysid shrimp population in the Scheldt estuary.\r\n\r\nPhase IV: Risk assessment\r\n\r\nIn the last phase, the results of the previous phases will be incorporated into an integrated risk assessment for the Scheldt estuary with reference to endocrine disrupting substances. Moreover, an overall risk assessment for the Scheldt estuary will be conducted with these data in correlation with environmental concentrations of the tested endocrine disrupters. Recommendations for sustainable management and priority substances will be developed.\r\n\r\nThe results of this study on \"endocrine disruption in the Scheldt estuary: distribution, exposure and effects\" will be used at different levels and in different ways.\r\n\r\nIn order to ensure (rapid) dissemination and valorisation of the obtained results, the following actions will be undertaken:\r\n\r\n- all results will be published in peer-reviewed journals;\r\n- transfer (and discussion) of the obtained knowledge and insights to national and international representatives of policy-making and policy-supporting governmental services. This discussion will offer an essential contribution to the international contractual engagements of Belgium;\r\n- diffusion of data and results through scientific databases;\r\n- interactive communication forum during the project (project website);\r\n- the results of the project will be brought together in a book, which will include the datasets on CD-rom and the main research results.\r\nThe diffusion and valorisation of the information as described above will be developed and maintained by the Flemish Institute for the Sea (VLIZ) (subcontracted) under supervision of the co-ordinator.","AbstractOtherLang":"<ul><li>Context\r\n\r\nEr is een groeiende bezorgdheid omtrent de mogelijkheid van sommige chemische stoffen om wijzigingen in het endocrien systeem bij mens en dier te veroorzaken. Zowel natuurlijke als nog betrekkelijk ongekende milieuverontreinigende stoffen blijken de hormoonhuishouding van mens en dier zodanig te kunnen verstoren dat nadelige effecten op de ontwikkeling en voortplanting kunnen optreden. \r\n\r\nDe eerste aanwijzingen van mogelijke effecten van endocrien verstorende stoffen en de aanwezigheid van deze verbindingen in het Schelde estuarium zijn recent gepubliceerd. De Gentse kanaalzone en het Antwerps industriegebied zijn er verantwoordelijk voor een groot deel van de huidige vervuiling. Er is dan ook een duidelijke nood aan onderzoek naar het optreden van hormoonverstoring in het Schelde estuarium. Bovendien is een grondige kennis van de verspreiding en langetermijneffecten van deze stoffen noodzakelijk in het kader van toekomstgerichte beleidsmaatregelen op nationaal en Europees niveau.\r\n\r\n\r\n<li>Beschrijving van het project\r\n\r\n<ul><li>Doelstellingen\r\n\r\nDe belangrijkste doelstellingen binnen het project zijn:\r\n\r\n- analyses van hormoonontregelaars in water, zwevende stof, sediment en biota (mysidacee\u00ebn, grondels);\r\n- in vitro evaluatie van de oestrogene en androgene potentie van de water, zwevende stof -en sedimentstalen;\r\n- ecotoxicologische en in situ evaluatie van de effecten op de brakwater-endeme mysidacee\u00ebn populatie (korte, middellange en lange termijn);\r\n- risicoschatting van hormoonontregelaars voor het Schelde estuarium.\r\n\r\n<li>Methodologie\r\n\r\nHet voorgestelde project kan worden ingedeeld in vier verschillende onderzoeksfasen:\r\n\r\nFase I: Verspreiding van hormoonontregelende stoffen in het Schelde estuarium (chemische analyses, analyses op biota, in vitro analyses)\r\n\r\nHet Schelde estuarium zal gedurende de duur van het project drie maal per jaar bemonsterd worden (lente, zomer en winter) voor het verzamelen van sediment, water, zwevende stof en biota (mysidacee\u00ebn en grondel). Een uitgebreide lijst van natuurlijke hormonen en hormoonverstorende chemicali\u00ebn zullen in deze stalen geanalyseerd worden.\r\nDe verschillende water, sediment en zwevende stof extracten worden eveneens in vitro getest op hun potentie om te binden met de humane oestrogeen en androgeenreceptor. De resultaten van deze analyses laten toe de oestrogene en androgene potentie in kaart te brengen in de verschillende milieumatrices van het Schelde estuarium.\r\n\r\nFase II: Evaluatie van de blootstelling van biota uit het Schelde estuarium aan hormoonontregelende stoffen\r\n\r\nUitgaande van de resultaten van de eerste bemonsteringscampagnes worden prioritaire verbindingen voor het Schelde estuarium aangeduid. Met de weerhouden hormoonverstoorders worden laboratorium experimenten opgezet met mysidacee\u00ebn om de acute en chronische effecten te evalueren (Fase III). \r\n\r\nFase III: Ecotoxicologische evaluatie van de effecten van hormoonontregelende stoffen die voorkomen in het Schelde estuarium op de endogene mysidacee\u00ebn populatie\r\n\r\nAcute en chronische mysidacee\u00ebn testen in het laboratorium \r\nOm de mogelijke effecten van de polluenten weerhouden in Fase II op mysidacee\u00ebn te evalueren worden acute en chronische ecotoxiciteittesten opgezet in het laboratorium. Mysidacee\u00ebn zullen eveneens worden blootgesteld in een multigeneratie-experiment over 2 generaties om de effecten van de hormoonverstoorder op lange termijn in te schatten. \r\n\r\nPopulatiestudie van de brakwater-endeme mysidacee\u00ebn in het Schelde estuarium\r\nDe mysidacee\u00ebn populatie van het Schelde estuarium wordt opgevolgd gedurende \u00e9\u00e9n jaar. Op deze manier kan de huidige populatie vergeleken worden met de beschikbare historische data en kunnen eventuele verschuivingen worden vastgesteld. Bovendien zal gebaseerd op de resultaten van de toxiciteittesten in het laboratorium, een meer uitgebreide effectstudie gebeuren in het veld. Gevoelige eindpunten in het laboratorium zullen eveneens getoetst worden in situ op de brakwater-endeme mysidacee\u00ebn populatie in het Schelde estuarium.\r\n\r\nFase IV: Risicoschatting \r\n\r\nIn een laatste fase zullen de resultaten van de voorgaande fasen gebundeld worden om tot een ge\u00efntegreerde risico-analyse te komen voor het Schelde estuarium met betrekking tot de problematiek van hormoonontregelaars. Bovendien zal uitgaande van deze gegevens, een globale risicoschatting gebeuren voor het Schelde estuarium met betrekking tot de aanwezige milieuconcentraties van de geteste hormoonontregelaars. Aanbevelingen voor duurzaam beheer en prioritaire probleemstoffen zullen worden aangeduid.\r\nDe resultaten van deze studie \u2018Endocriene verstoring in het Schelde estuarium: verspreiding, blootstelling en effecten\u2019 zullen op verschillende manieren en op verschillende niveaus gebruikt worden. \r\nOm doorvloei en valorisatie van de bekomen kennis te verzekeren worden volgende acties ondernomen:\r\n\r\n- alle resultaten worden gepubliceerd in (inter)nationale tijdschriften met lezerscomit\u00e9;\r\n- overdracht (en discussie) van de bekomen resultaten en inzichten met vertegenwoordigers van nationale en internationale beleid- en beleidsondersteunende instanties, waardoor een essenti\u00eble bijdrage geleverd wordt aan het vervullen van de internationale contractuele engagementen van Belgi\u00eb;\r\n- verspreiden van de gegevens en resultaten via wetenschappelijke databanken;\r\n- interactief communicatieforum tijdens het project (project website);\r\n- de resultaten van het project zullen gebundeld worden in een boek, incl. datasets op CD-ROM, met de belangrijkste resultaten van het onderzoek.\r\nDe doorvloei en valorisatie van de informatie zoals hierboven beschreven zal ontwikkeld en onderhouden worden door het Vlaams Instituut voor de Zee (VLIZ) in onderaanneming en onder opvolging van de
