Amongst the potentially toxic representatives, P. pungens is the most abundant in the North Sea. \r\n\r\nThe goal of this study is to gain a better understanding of the patterns in genetic, reproductive  ecophysiological and morphological variation and the relationship to toxin production within this \"species\". This will lead to a better insight in the distribution of potentially toxic strains within these species and will help developing efficient detection methods. \r\n\r\nWe are building a collection of monoclonal cultures of Pseudo-nitzschia pungens from different stations in the North Sea and adjoining areas (estuaries and coastal lagoons), and other parts of the world. In order to elucidate the sexual compatibility between different strains, crossing experiments are being carried out. Potential genetic differentiation between allo- and sympatric populations and their phylogenetic relationships are assessed using the internal transcribed spacers (ITS1 and ITS2) of the nuclear ribosomal DNA, which has previously been successfully used for phylogenetic analyses in Pseudo-nitzschia species complexes. Some of these studies have demonstrated the presence of genetically distinct but morphologically indistinguishable species.","AbstractOtherLang":"Het mariene, planktonische diatomeeëngenus Pseudo-nitzschia omvat verschillende soorten die het toxine domoic acid kunnen produceren. Dit toxine veroorzaakt Amnesic Shellfish Poisoining bij mensen en hogere diersoorten. Van de verschillende potentieel toxische vertegenwoordigers is het P. pungen/multiseries complex het meest abundant in de Noordzee. Het doel is een beter inzicht te verwerven in de genetische, reproductieve en morfologische variatiepatronen in dit complex. Dit zal bijdragen tot een beter inzicht in de distributie van potentieel toxische stammen in dit complex en kan helpen bij het ontwikkelen van efficiente detectiemethodes. Om dit doel te bereiken wordt een collectie opgebouwd van monoklonale culturen van P. pungen en P. multiseries afkomstig van de Noordzee, de Westerschelde en de Spuikom van Oostende. De bedoeling is deze collectie uit te breiden met stammen van overal ter wereld. Aan de hand van kruisingsexperimenten wordt de seksuele compatibiliteit tussen verschillende stammen bestudeerd. Hiermee kunnen cryptische soorten gedetecteerd worden en de identificaties op basis van morfologie gevalideerd worden. De genetische differentiatie en fylogenetische verwantschappen zullen geanalyseerd worden op basis van de internal transcribed spacers (ITS1 and ITS2) van het nucleair ribosomaal DNA.
