Lysosomal hydrolases are associated with these granules in both organisms, indicating that they are lysosomes. Cytophotometry has been used to determine the quantitative effects of Cu<sup>2+<\/sup> , Cd<sup>2+<\/sup> , and Hg<sup>2+<\/sup> on the cytochemical staining reaction for N-acetyl-B-D-hexosaminidase in<i> C. flexuosa<\/i>. These metal ions significantly increased the staining reaction of hexosaminidase with increasing concentrations, and the response thresholds are comparable to known environmental levels. Pretreatment of colonies with Cu<sup>2+<\/sup> gave no indication of tolerance adaptation, although there is evidence of the cumulative toxicity of Cu<sup>2+<\/sup>. Hydrocortisone reduced the staining intensity of free hexosaminidase in Cu<sup>2+<\/sup>-treated animals, indicating that the toxic effect may involve decreased stability of lysosomal membranes.
