The most pronounced problem is that one measures on the system-level whereas the results consist of the combined effect of (1) the physiological properties of the different algal species and (2) the interactions between autotrophs and heterotrophs. Excretion turned out to be negligible. In order to have a control on the existence of \"bottle-enclosure effects\", a series of short incubations was carried out simultaneously with a long incubation. Over the same time the production of the short incubations was less than that of the one long incubation. This may be caused by a temporal decrease of the rate of production at the beginning of the incubation. A third factor, the influence of the bacterioplankton on <sup>14</sup>C results, could not be investigated because of methodological problems. For measuring the primary production on the species level autoradiography seemed a suitable method. When using a light microscope two variants are possible: grain density and track- autoradiography. For quantative application track-autoradiography should be the method of choice because there is no relevant background, which makes interpretation of the auto- radiograms more reliable. In practice this method proved to be a difficult one. The problems encountered are discussed in detail. Primary production estimates from track-counts agreed well with those calculated from carbon content of the cell and doubling-time.","AbstractOtherLang":"Aan het meten van de primaire productie van aquatische oecosystemen met behulp van de <sup>14</sup>C-methode kleeft een aantal bezwaren. Het voornaamste is wel dat de methode meet op systeem- niveau terwijl de resultaten de opstelsom zijn van (1) de fysiologische eigenschappen van de verschillende algen-soorten en (2) de interacties tussen autotrophen en heterotrophen. Aan drie van dit type factoren is enig onderzoek verricht. De eerste, excretie, bleek verwaarloosbaar. De invloed van de incubatiemethode, met het oog op \"bottle-enclosure-effecten\", werd onderzocht door vergelijking van een serie korte incubaties bij elkaar, met één lange. De productie van de eerste serie was langer dan die van de lange incubatie. Wellicht wordt dit veroorzaakt door een tijdelijke terugslag in opnamesnelheid aan het begin van een incubatie. Onderzoek naar de invloed van bakteriën stuitte op methodische problemen. De bepaling van de primaire productie op (cel-c.q.) soortsniveau bleek mogelijk met autoradiografie. Van de voor het lichtmicroscoop geschikte varianten van deze methode verdient track-autografie bij kwantitatieve bepaling de voorkeur boven grain density-autoradiografie, omdat bij de eerste variant relevante achtergrond vrijwel ontbreekt, hetgeen correcte interpretatie van de autoradiogrammen belangrijk eenvoudiger maakt. De practische uitvoering van track-autoradiografie bleek niet eenvoudig; de ondervonden problemen worden in detail besproken.
