In order to fill in this gap this study was started. Special attention was paid to finding the best method for measuring <i>in situ</i> primary production. The <sup>14</sup>C-method proved to be superior to the O<sub>2</sub>-method, because of the frequent appearance of air bubbles in the oxygen bottles caused by warming up of the bottles during incubation on the tidal flat. Dry combustion and subsequent collection of the <sup>14</sup>CO<sub>2</sub> in a mixture of ethanolamine and 2-methoxyethanol gave the best results if the algae had to be prepared for liquid-scintillation-counting. This counting was carried out in a mixture of toluene (containing P.P.O. and P.O.P.O.P.) and 2-methoxyethanol and gave efficiency-values up to 80%. Comparison of the <sup>14</sup>C-method with tbe O<sub>2</sub>-method (Gilson Respirometer) gave comparable results.
