These studies require a sensitive, accurate, rapid, and reliable technique for genetic sexing of eggs, larvae, and adults. In this study, we report a simplex polymerase chain reaction approach that uses a single pair of primers for simultaneous amplification of sex-specific amplicons. Males and females yield a single diagnostic band of 933 and 1,906 bp, respectively, in three different strains of medaka tested, permitting gender identification accurately of both immature and adult fish.
