Unusually, <i>M. acetivorans</i> Pgb (<i>Ma</i>Pgb) binds preferentially O<sub>2</sub> rather than CO and displays anticooperativity in ligand binding. Here, kinetics and/or thermodynamics of ferric and ferrous <i>Ma</i>Pgb (<i>Ma</i>Pgb(III) and <i>Ma</i>Pgb(II), respectively) nitrosylation are reported. Data were obtained between pH 7.2 and 9.5, at 22.0 °C. Addition of NO to <i>Ma</i>Pgb(III) leads to the transient formation of <i>Ma</i>Pgb(III)–NO in equilibrium with <i>Ma</i>Pgb(II)–NO<sup>+</sup>. In turn, <i>Ma</i>Pgb(II)–NO<sup>+</sup> is converted to <i>Ma</i>Pgb(II) by OH<sup>−</sup>-based catalysis. Then, <i>Ma</i>Pgb(II) binds NO very rapidly leading to <i>Ma</i>Pgb(II)–NO. The rate-limiting step for reductive nitrosylation of <i>Ma</i>Pgb(III) is represented by the OH<sup>−</sup>-mediated reduction of <i>Ma</i>Pgb(II)–NO<sup>+</sup> to <i>Ma</i>Pgb(II).
