Title

Uji aktivitas enzim dan patogenisitas bakteri simbion Sargassum polycystum penghasil alginat lyase Cytobacillus kochii pada udang vaname (Litopenaeus vannamei)

Abstract

Cytobacillus kochii is a Sargassum symbiont bacterium from Lampung waters that has alginate lyase activity to hydrolyze alginate into oligosaccharides that have potential as immunostimulants in shrimp farming. The purpose of this study was to test the alginate lyase enzyme activity and pathogenicity of alginate lyase producing symbiont bacteria S. polycystum C. kochii on Pacific white shrimp. This research was conducted at the Laboratory of Aquaculture, Department of Fisheries and Marine, Faculty of Agriculture, University of Lampung. The calculation of alginate lyase activity was carried out by giving Cetylpyridinium chloride (CPC) solution at a concentration of 0.8% and 1% with an incubation period of 24, 48 , and 72 hours. Patogenicity test was desain used completely randomized design (CRD) with 5 treatments bacterial density 0, 102, 103, 104, and 106 CFU/mL with 3 replications. The treatment gave to Pacific white shrimp with an initial average weight of 3 ± 0 grams. Clinical symptoms and mean time to death were observed for 7 days of rearing. The results showed that the highest alginate lyase activity was obtained with a clear zone value of 6.20 mm in 0.8% CPC solution with an incubation time of 72 hours. Meanwhile, the LD50 value was not reached because up to 7 days there was only 1 death of shrimp with a survival rate value 90% and an average time of death of 56 hours after infection. The conclusion of the research is that the optimum incubation time to produce alginate lyase enzyme is 0.8% CPC concentration with 72 hours incubation time and it is suspected that C. kochii bacteria are not pathogenic to Pacific white shrimp, but have the potential to cause disease in a longer time.
