Title
DNA barcoding reveals high diversity of aquatic invertebrates in Setiu Wetlands, Malaysia
Abstract
Setiu Wetlands is a coastal wetland located in east Peninsular, Malaysia known for its productivity and rich biodiversity. However, the diversity of aquatic invertebrates in this unique ecosystem remains poorly understood. This study examined the diversity of aquatic invertebrates in the Setiu Wetlands by analysing 98 DNA sequences representing four classes, nine orders, and 15 families using the cytochrome c oxidase subunit I (COI) barcode region. DNA barcoding analysis and automatic species delimitation uncovered 32 Molecular Operational Taxonomic Units (MOTUs). Gastropods and malacostracans exhibited the highest species richness, followed by bivalves and merostomata. DNA barcoding provided valuable insights into the phylogenetic relationships among taxa and identified potentially cryptic species within the presumed mopho-species, Cassidula nucleus and Penaeus monodon. Our findings demonstrate the effectiveness of DNA barcoding for rapidly assessing invertebrate diversity in these understudied wetlands. By constructing a barcode library for the Setiu Wetlands, this research provides important data for future conservation studies of aquatic invertebrates. This barcode data will also enable more efficient species identification in future research, aiding in the monitoring of biodiversity and ecosystem health over time.
